TI - Efferocytosis of SARS-CoV-2-infected dying cells impairs macrophage anti-inflammatory programming and continual clearance of apoptotic cells AID - 10.1101/2021.02.18.21251504 DP - 2021 Jan 01 TA - medRxiv PG - 2021.02.18.21251504 4099 - http://medrxiv.org/content/early/2021/02/23/2021.02.18.21251504.short 4100 - http://medrxiv.org/content/early/2021/02/23/2021.02.18.21251504.full AB - COVID-19 is a disease of dysfunctional immune responses, but the mechanisms triggering immunopathogenesis are not established. The functional plasticity of macrophages allows this cell type to promote pathogen elimination and inflammation or suppress inflammation and promote tissue remodeling and injury repair. During an infection, the clearance of dead and dying cells, a process named efferocytosis, can modulate the interplay between these contrasting functions. Here, we show that engulfment of SARS-CoV2-infected apoptotic cells (AC) exacerbates inflammatory cytokine production, inhibits the expression of efferocytic receptors, and impairs continual efferocytosis by macrophages. We also provide evidence that monocytes from severe COVID-19 patients express reduced levels of efferocytic receptors and fail to uptake AC. Our findings reveal that dysfunctional efferocytosis of SARS-CoV-2-infected cell corpses suppress macrophage anti-inflammation and efficient tissue repair programs and provide mechanistic insights for the pathogenesis of the hyperinflammation and extensive tissue damage associated with COVID-19.Competing Interest StatementThe authors have declared no competing interest.Funding StatementThis work was supported by grants from the Fundo de Amparo a Pesquisa do Estado de Sao Paulo (FAPESP, 2018/255594 and 2020/052886), Coordenacao de Aperfeicoamento de Pessoal de Nivel Superior (CAPES 88887.507253/202000) and Conselho Nacional de Desenvolvimento Cientifico e Tecnologico (CNPq, 434538/20183)Author DeclarationsI confirm all relevant ethical guidelines have been followed, and any necessary IRB and/or ethics committee approvals have been obtained.YesThe details of the IRB/oversight body that provided approval or exemption for the research described are given below:The procedures followed in the study were approved by the Research Ethics Committee of Hospital das Clinicas de Ribeirao Preto (CEP-FMRP/USP) and by the National Ethics Committee, Brazil (Comissao Nacional de Etica em Pesquisa (CONEP), protocols 30248420.9.0000.5440 and 39722020.9.0000.5440.All necessary patient/participant consent has been obtained and the appropriate institutional forms have been archived.YesI understand that all clinical trials and any other prospective interventional studies must be registered with an ICMJE-approved registry, such as ClinicalTrials.gov.
